RESUMO

The aim of this study was to develop and characterize a double layer biomembrane for dual drug delivery to be used for the treatment of wounds. The membrane was composed of chitosan, hydroxypropyl methylcellulose and lidocaine chloride (anesthetic drug) in the first layer, and of sodium alginate-polymyxin B sulphate (antibiotic) nanoparticles as the second layer. A product with excellent thickness (0.01-0.02 mm), adequate mechanical properties with respect to elasticity, stiffness, tension, and compatible pH for lesion application has been successfully obtained. The incorporation of the drugs was confirmed analysing the membrane cross-sections by scanning electron microscopy. A strong interaction between the drugs and the functional groups of respective polymers was confirmed by Fourier-Transform Infrared Spectroscopy, thermal analysis and X-ray diffraction. Microbiological assays showed a high antimicrobial activity when polymyxin B was present to act against the Staphylococcus aureus and Pseudomonas aeruginosa strains. Low cytotoxicity observed in a cell viability colorimetric assay and SEM analysis suggest biocompatibility between the developed biomembrane and the cell culture. The in vivo assay allowed visualizing the healing potential by calculating the wound retraction index and by histological analysis. Our results confirm the effectiveness of the developed innovative biomaterial for tissue repair and regeneration in an animal model.

Release of engineered nanoparticles (ENPs) to soil is well documented but little is known on the subsequent soil-pore water distribution of ENPs once present in soil. In this study, the availability and mobility of silver (Ag) and gold (Au) ENPs added to agricultural soils were assessed in two separate pot experiments. Pore water samples collected from pots from day 1 to 45 using porous (<0.17 µm) membrane samplers suggest that both Ag and Au are retained almost completely within 24 h with less than 13% of the total added amount present in pore water on day 1. UV-Vis and TEM results showed that AuENPs in pore water were present as both homoaggregates and heteroaggregates until day 3 after which the concentration in pore water was too low to detect the presence of aggregates. A close relation between the concentration of Au and Fe in pore water suggests that the short term solubility of Au is partly controlled by natural soil colloids. Results suggest that under normal aerated soil conditions the actual availability of Ag and AuENPs is low which is relevant in view of risk assessment even though the impact of environmental conditions and soil properties on the reactivity of ENPs (and/or large ENPs aggregates) retained in the solid matrix need to be addressed further.

The sorption capacity of nanoporous titanosilicate Engelhard titanosilicate number 4 (ETS-4) and silica-coated magnetite particles derivatised with dithiocarbamate groups towards Hg(II) was evaluated and compared in spiked ultra-pure and spiked surface-river water, for different batch factors. In the former, and using a batch factor of 100 m(3)/kg and an initial Hg(II) concentrations matching the maximum allowed concentration in an effluent discharge, both materials achieve Hg(II) uptake efficiencies in excess of 99 % and a residual metal concentration lower than the guideline value for drinking water quality. For the surface-river water and the same initial concentration, the Hg(II) uptake efficiency of magnetite particles is outstanding, achieving the quality criteria established by the Water Framework Directive (concerning Hg concentration in surface waters) using a batch factor of 50 m(3)/kg, while the efficiency of ETS-4 is significantly inferior. The dissimilar sorbents' Hg(II) removal efficiency is attributed to different uptake mechanisms. This study also highlights the importance of assessing the effective capacity of the sorbents under realistic conditions in order to achieve trustable results.

The Epstein-Barr virus (EBV) is the etiological agent of oral hairy leukoplakia (OHL), an oral lesion with important diagnostic and prognostic value in acquired immunodeficiency disease syndrome. The two EBV genotypes, EBV-1 and EBV-2, can be distinguished by divergent gene sequences encoding the EBNA-2, 3A, 3B, and 3C proteins. The purpose of this study was to identify the EBV genotype prevalent in 53 samples of scrapings from the lateral border of the tongue of HIV-1 seropositive patients, with and without OHL, and to correlate the genotypes with presence of clinical or subclinical OHL with the clinic data collected. EBV-1 and EBV-2 were identified through PCR and Nested-PCR based on sequence differences of the EBNA-2 gene. EBV-1 was identified in the 31 samples (15 without OHL, 7 with clinical OHL and 9 with subclinical OHL), EBV-2 in 12 samples (10 without OHL, 1 with clinical and 1 subclinical OHL), and a mixed infection in 10 samples (2 without OHL, 3 with clinical and 5 with subclinical OHL). The presence of EBV-1 was higher in women, but a significant statistical result relating one the EBV genotypes to the development of OHL was not found. We conclude that the oral epithelium in HIV-1 seropositive patients can be infected by EBV-1, EBV-2 or by a mixed viral population.

Between January and March 2001, an outbreak of jaundice and hemorrhagic fever occurred in the state of Minas Gerais, Southeast region of Brazil, in which a mortality rate of 53% was reported. Seroconversion, virus isolation, histopathological and immunohistochemical findings, and reverse transcription-polymerase chain reaction (RT-PCR) identified yellow fever virus (YFV) as the etiological agent responsible for the outbreak. Partial nucleotide sequence analysis from a fragment of the YFV genome spanning parts of nonstructural (NS) 5 gene and 3' noncoding region (3' UTR) showed that the YFV involved in this outbreak belongs to South American genotype I and differs from the Brazilian virus identified in 1996.

PURPOSE: To study the spatial arrangement of the bundles of myocardial fibers presents in the left atrial-venous junctions and in the wall of the pulmonary veins. METHODS: The study was made on 24 human adult hearts, together with pulmonary vessels, fixed in 10% formaldehyde solution. Each specimen was cleared of remnants of pericardium to expose the myocardial fibers as clearly as possible. Particular attention was paid to the atrial-venous junction and the extension of the myocardial fibers in the pulmonary veins. The specimens were embedded in celloidin and cut serially at a thickness of 70 mu and stained by methods of Azan's trichrome. RESULTS: The myocardial bundles leave the atrial wall around the openings of all pulmonary veins forming a sphincter-like structure. These fibers are continuous in the adventitial coat running in oblique or spiral directions. Some bundles, more external, leaving from the atrial wall, surround the ostium of the veins and return to the atrium, forming true loops. CONCLUSION: The extremities of the pulmonary veins and their junctions with the atrium have a morphological substract which may be of physiological importance in the control of the pulmonary venous pressure and blood flow.
